CJC-1295 (NO DAC) 5mg + Ipamorelin 5mg Vial

This vial combines two synthetic peptides that influence growth-hormone release through separate receptors. Each is present at 5mg, giving 10mg of freeze-dried peptide in total.

CJC-1295 without DAC (Mod GRF 1-29)

The first component is a modified copy of the active 29-residue core of growth hormone-releasing hormone (GHRH). Four amino-acid substitutions (D-Ala2, Gln8, Ala15 and Leu27) make it more resistant to the enzymes that clear native GHRH within minutes. It has no Drug Affinity Complex: the maleimidopropionic acid linker that lets the better-known CJC-1295 with DAC attach to albumin and stay in circulation for days. Without that tether it behaves as a short-acting signal at the GHRH receptor, with a half-life described as being in the region of 30 minutes.

Ipamorelin

The second component is a pentapeptide, Aib-His-D-2-Nal-D-Phe-Lys-NH2, developed at Novo Nordisk under the code NNC 26-0161. It is not a GHRH analog. It activates GHS-R1a, the receptor later shown to be the natural target of ghrelin, which is why it is often described as a ghrelin mimetic.

Why the label says "no DAC"

The name CJC-1295 is used for both molecules, and much of the published human data on CJC-1295, including the multi-day rises in GH and IGF-1 and the 5.8 to 8.1 day half-life reported in 2006, was generated with the DAC form. A fixed 1:1 blend under one seal

Both peptides are freeze-dried together in a single crimp-sealed glass vial, so a study that needs the pairing works from one lot instead of two separate products. Lyophilised powder also keeps far better in storage than peptide held in solution.

Each component named on its own

The label states CJC-1295 (no DAC) and ipamorelin with their own 5mg quantities, so an analytical lab can match each one by name and content. Growth-hormone release from the pituitary is controlled from two directions. GHRH acts at its own receptor on somatotroph cells, while ghrelin-type signals act at GHS-R1a. Because the receptors differ, the two inputs can add to each other instead of competing for the same site.

The physiology behind the pairing

In a 2001 human physiology study, Arvat and colleagues gave ghrelin and GHRH together and reported a growth-hormone response larger than either hormone produced alone. That work used the natural hormones, not this blend, so it supports the idea of complementary signalling rather than any claim about a commercial vial.

What is known about ipamorelin

Raun et al. (1998) called ipamorelin the first selective growth-hormone secretagogue. In conscious swine it released GH without raising ACTH or cortisol beyond the levels seen with a GHRH stimulus, and without changing prolactin, which set it apart from GHRP-2 and GHRP-6. Its effect was blocked by a GHRP-receptor antagonist but not by a GHRH antagonist. A 1999 intravenous study in healthy men reported a terminal half-life of about two hours and a single GH peak at roughly 40 minutes.

What is known about Mod GRF 1-29

Published data on the no-DAC molecule are much thinner than for the DAC version. Its profile is mostly inferred from structure: the substitutions slow enzymatic breakdown, giving a brief GHRH-receptor signal that sits closer to native GHRH(1-29) than to the albumin-bound form.

Where the evidence stops

No mature peer-reviewed human trial of a CJC-1295 + ipamorelin blend has been published. The one registered efficacy trial of ipamorelin, a 2014 Phase 2 study in post-operative ileus, found no significant benefit over placebo, and development did not continue.
