Thymosin beta-4 (Tβ4) is a 43-amino-acid peptide found in most mammalian cells, where it acts as the main buffer for monomeric G-actin. It binds G-actin one-to-one and helps set how much actin is free to build filaments, which shapes how cells move and change shape.

TB-500 is the market name for something smaller. Analytical and doping-control chemistry identifies the compound sold under this label as a short acetylated synthetic fragment, Ac-LKKTETQ, taken from the actin-binding region of Tβ4. That matters when you read the literature: most published work, and every human trial on record, used the full-length protein. Findings from Tβ4 papers describe a related molecule, not this vial.

What the Preclinical Literature Reports

Most of the evidence comes from cell culture and animal models, much of it using full-length Tβ4 rather than the fragment. Short actin-binding fragments have shown activity in preclinical models, but results for one molecule should not be assumed for the other.

Actin and cell movement

By controlling the pool of free G-actin, Tβ4 influences lamellipodial protrusion and the migration of fibroblasts and keratinocytes. These are the cell behaviours that close a wound surface, which is why actin work anchors this research area.

New vessel growth

In-vitro and rodent studies report that Tβ4 encourages endothelial cells to migrate and form tube-like structures, with collateral vessel growth in ischaemic tissue. This is one of the more often replicated signals in the field.

Heart, eye and skin injury models

A 2004 mouse study after induced myocardial infarction linked Tβ4 to cardiomyocyte survival through integrin-linked kinase signalling. Rabbit and rodent corneal models show faster re-epithelialisation, and rat and diabetic-mouse skin wounds closed faster in published work, including one arm testing an actin-binding-domain peptide.

The AcSDKP side-story

The N-terminal tetrapeptide of Tβ4, AcSDKP, is studied separately as an anti-fibrotic and haematopoietic regulator. Claims that bundle this activity under the TB-500 name are mixing two distinct pieces of biology.
